Radiometric Assay

As a relatively simple but extremely powerful tool for studying receptors and ion channels, radioligand binding assay is widely employed to analyze interactions of hormones, neurotransmitters, grow factors and related drugs with the receptors/ion channels. At HDB we built the capacity of performing HTS-compatible large scale radioligand binding assay screening for all the GPCRs and specific ion channels from our cell lines list at low costs. We have optimized protocols for both ligand binding assays such as hERG binding and GTPgammas binding such as opioid receptors and group II and III mGluRs. Moreover with advanced Toshiba high-speed refrigerated centrifuges and IKA homogenizers, we built strong membrane preparation capacity targeting numerous GPCRs at a much lower price compared to other vendors. The radiometric assays can also be used in kinase assays using 32P-ATP or PDE assays using 3H-cAMP or 3H-cGMP.

Radiometric Assay Facility
With MicroBeta Trilux and cell harvester facility, we have built the capability of performing assays such as ion channel/ GPCR ligand binding, kinase assay, GTPgammaS binding, cell uptake assays, cytotoxicity assays etc using 3H, 14C, 32P, 33P, 35S, 51Cr, 125I or glow-type luminescence labels at HDB. The MicroBeta-compatible 96-well filters/microplates and 384-well microplates make large-scale compound screening applicable.

1450 MicroBeta TriLux is a 12-detector instrument designed for liquid scintillation or luminescence detection of samples in microplates, tubes or on filters. Controlled by windows work station software, by combining LSC reliability with plate reader simplicity, it allows vast savings in time, consumables and waste.

Cell Harvester Facility
The cell harvester from Perkin Elmer is critical facility for assays such as cell proliferation assays using tritiated thymidine uptake, receptor binding assays, adherent cell assays, cytotoxicity assays and nucleic acid degradation.

Toshiba Hitachi High-Speed Refrigerated Centrifuge
HDB has Toshiba Hitachi high-speed refrigerated centrifuge model CR22GII. The centrifuge has a motor which can hold 6 x50ml centrifuge tubes and can reach maximal speed of 22,000 rpm. This brand-new centrifuge makes the critical high-speed centrifugation process during membrane preparation much easier here at HDB.
