| Sample Case: on one Thursday, project manager from the client instructs compound manager at its chemistry service provider to ship about 1,000 compounds to HDB. The received compounds will be mixed with singletons and a focused library. The shipment is delivered to HDB on Friday and HDB compound manager immediately notifies prospective HDB project manger to initiate hit-to-lead process. The project team starts conducting biochemical and cell-based assays the following Monday. Within 5 working days, all the data generated within the week including cross-plate triplicate IC50s are deposited into client’s project files. Based on this information, a new instruction of cherry picking of selective compounds, selectivity profiling and in vitro ADME tests is sent to HDB by the client. All these data are used to support lead optimization. |

| The structure of most ultimately marketed drugs is derived from early leads. The process leading from leads to the selection of development candidates is a critical step in drug discovery. The Hit-to-Lead study is the follow up step after high-throughput screening (HTS), and challenging coordination of efforts in chemistry, biology, and pharmacology. By using HDB’s Hit-to-Lead contract discovery services, the compounds, either singleton or focused library, if they are synthesized by chemistry CROs locally, do not need to be shipped back to clients’ site before bioactivity data can be obtained. This work flow remarkably reduces turn around time for lead discovery as well as the cost. |
